What is the right way to check the 400 oil level?

A really basic question - to check the oil level is this done with the dipstick resting on the casing thread or fully screwed back in?

Honda CB500 K1 (new pit dug out ready)

Dip stick resting on the top of the opening with the bike on the centre stand.
